The microscope allows the observation of single cells, the building blocks of life. Since its invention, the resolution, and particularly the contrast, of the instrument has been continually refined. During most of its evolution, however, the microscopist has remained a passive observer. With the recent development of optical manipulation techniques, this situation has completely changed.
